Try them out in friendlies and see what seems to work well.

There are some "well known" attributes that contribute to good central defending. You need tackling, marking, heading; strength; decisions, determination, concentration. Your FM tactics setups will show you the ones highlighted for the style of defender you choose (defend, stopper, cover). You don't have to - and for a lower league side you can't expect - to have players who are good at all of these. In general, it's probably better to have decent ratings at all of them than great at something and really terrible at something (especially don't let in a player with poor concentration: that's the one that lets you play well a lot of the time but then occasionally make disastrous blunders). In past years, FM would punish you quite severely if you didn't have at least one defender who is very good in the air, or opponents will just pump the ball into the box and score bunches on you, usually from corners. Not sure if FM12 is as severe about this.
